Daisy Werthan
Daisy is difficult to get along with, to say the least. Like many people who grew up in the Old South, change is hard for her. Most of the film is a battle between Hoke and Daisy to simply get her to let him do his job. It isn't necessarily dramatic, but it's enough of a conflict to make her the antagonist for the first half of the film.
You could also say that general racism and prejudice is the antagonist of the movie, but Miss Daisy embodies that, even if she doesn't do so purposefully or maliciously.
